Genes affected
APC (HGNC:583): (APC regulator of WNT signaling pathway) This gene encodes a tumor suppressor protein that acts as an antagonist of the Wnt signaling pathway. It is also involved in other processes including cell migration and adhesion, transcriptional activation, and apoptosis. Defects in this gene cause familial adenomatous polyposis (FAP), an autosomal dominant pre-malignant disease that usually progresses to malignancy. Mutations in the APC gene have been found to occur in most colorectal cancers, where disease-associated mutations tend to be clustered in a small region designated the mutation cluster region (MCR) and result in a truncated protein product. Pathogenic, criteria provided, single submitterclinical testingMyriad Genetics, Inc.May 03, 2023This variant is considered pathogenic. This variant creates a frameshift predicted to result in premature protein truncation. -
Pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpDec 18, 2023This sequence change creates a premature translational stop signal (p.Asn627Leufs*2) in the APC gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in APC are known to be pathogenic (PMID: 17963004, 20685668). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with familial adenomatous polyposis (PMID: 8381580, 9067764, 19029688, 20685668). This variant is also known as 1879del4. ClinVar contains an entry for this variant (Variation ID: 411536). RNA analysis performed to evaluate the impact of this premature translational stop signal on mRNA splicing indicates it does not significantly alter splicing (Invitae). For these reasons, this variant has been classified as Pathogenic. -

Pathogenic, criteria provided, single submitterclinical testingAmbry GeneticsApr 05, 2017The c.1879_1882delAACA pathogenic mutation, located in coding exon 14 of the APC gene, results from a deletion of 4 nucleotides at nucleotide positions 1879 to 1882, causing a translational frameshift with a predicted alternate stop codon (p.N627Lfs*2). This alteration has been previously reported in an Italian familial adenomatous polyposis (FAP) family (Scarano MI et al. Hum. Mutat., 1997;9:191-3). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
