It is also involved in other processes including cell migration and adhesion, transcriptional activation, and apoptosis. The p.E1209* variant (also known as c.3625G>T), located in coding exon 15 of the APC gene, results from a G to T substitution at nucleotide position 3625. This changes the amino acid from a glutamic acid to a stop codon within coding exon 15. This alteration occurs at the 3' terminus of theAPC gene, is not expected to trigger nonsense-mediated mRNA decay, and only impacts the last 1635 amino acids of the protein. However, premature stop codons are typically deleterious in nature and the impacted region is critical for protein function and a significant portion of the protein is affected (Ambry internal data). Other truncating alterations downstream have been observed in individuals with a personal and/or family history that is consistent with APC-related disease (Ambry internal data). Based on the supporting evidence, this alteration is interpreted as a disease-causing mutation.
