The p.S2555* variant (also known as c.7664C>A), located in coding exon 15 of the APC gene, results from a C to A substitution at nucleotide position 7664. This changes the amino acid from a serine to a stop codon within coding exon 15. This alteration occurs at the 3' terminus of the APC gene, is not expected to trigger nonsense-mediated mRNA decay, and only impacts approximately 10% of the protein. However, premature stop codons are typically deleterious in nature, and structural analysis suggests this deletion removes a known motif (Thr2841-Ser2842-Val2843) needed for protein binding involved in regulation of protein function (Slep KC et al, PLoS ONE 2012; 7(11):e50097; Zhang Z et al, PLoS ONE 2011; 6(8):e23507). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
