Uncertain significance, no assertion criteria provided | clinical testing | Department of Pathology and Laboratory Medicine, Sinai Health System | - | The APC p.Asp2656Tyr variant was identified in 1 of 1260 proband chromosomes (frequency: 0.001) from individuals or families with sproradic colorectal cancer (Christie 2013). The patient identified in this study was found to have a second APC variant c.3878delC. The patient tumour showed loss of heterozygosity. The variant was also identified in the COSMIC database. The variant was not identified in dbSNP, NHLBI Exome Sequencing Project (Exome Variant Server), the Exome Aggregation Consortium, Clinvitae database, InSiGHT Colon Cancer Gene Variant Database (LOVD), Zhejiang Colon Cancer Database (LOVD), ClinVar database, GeneInsight - COGR database, and UMD. The p.Asp2656 residue is conserved across mammals and other organisms, and computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) suggest that the variant may impact the protein; however, this information is not predictive enough to assume pathogenicity.
