The c.2050A>T (p.I684F) alteration is located in exon 17 (coding exon 15) of the MEGF10 gene. This alteration results from a A to T substitution at nucleotide position 2050, causing the isoleucine (I) at amino acid position 684 to be replaced by a phenylalanine (F). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
MEGF10-related myopathy Uncertain:1
This sequence change replaces isoleucine with phenylalanine at codon 684 of the MEGF10 protein (p.Ile684Phe). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and phenylalanine. This variant is present in population databases (rs772306149, ExAC 0.005%). This variant has not been reported in the literature in individuals affected with MEGF10-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0").
