The p.K122N variant (also known as c.366G>C) is located in coding exon 4 of the RAD50 gene. The lysine at codon 122 is replaced by asparagine, an amino acid with similar properties. This change occurs in the first base pair of coding exon 4. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This sequence change replaces lysine, which is basic and polar, with asparagine, which is neutral and polar, at codon 122 of the RAD50 protein (p.Lys122Asn). This variant is present in population databases (rs587782384, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with RAD50-related conditions. ClinVar contains an entry for this variant (Variation ID: 142325). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. The frequency of this variant in the general population, 0.0000071 (2/282672 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign.
