The p.R278L variant (also known as c.833G>T), located in coding exon 6 of the RAD50 gene, results from a G to T substitution at nucleotide position 833. The arginine at codon 278 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 278 of the RAD50 protein (p.Arg278Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RAD50-related conditions. ClinVar contains an entry for this variant (Variation ID: 220537).
