The p.N758S variant (also known as c.2273A>G), located in coding exon 14 of the RAD50 gene, results from an A to G substitution at nucleotide position 2273. The asparagine at codon 758 is replaced by serine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 758 of the RAD50 protein (p.Asn758Ser). This variant is present in population databases (rs147568421, gnomAD 0.004%). This missense change has been observed in individual(s) with breast cancer (PMID: 35534704). ClinVar contains an entry for this variant (Variation ID: 232761). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt RAD50 protein function with a negative predictive value of 80%.
