Variant summary
Our verdict is Uncertain significance. The variant received 4 ACMG points: 4P and 0B. PM2PP3_Moderate
The NM_005732.4(RAD50):c.3640C>G(p.Arg1214Gly) variant causes a missense change. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. R1214C) has been classified as Uncertain significance.

The p.R1214G variant (also known as c.3640C>G), located in coding exon 24 of the RAD50 gene, results from a C to G substitution at nucleotide position 3640. The arginine at codon 1214 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
