The p.H1269Q variant (also known as c.3807T>G), located in coding exon 25 of the RAD50 gene, results from a T to G substitution at nucleotide position 3807. The histidine at codon 1269 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This sequence change replaces histidine, which is basic and polar, with glutamine, which is neutral and polar, at codon 1269 of the RAD50 protein (p.His1269Gln). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
