Genes affected
SH3TC2 (HGNC:29427): (SH3 domain and tetratricopeptide repeats 2) This gene encodes a protein with two N-terminal Src homology 3 (SH3) domains and 10 tetratricopeptide repeat (TPR) motifs, and is a member of a small gene family. The gene product has been proposed to be an adapter or docking molecule. Mutations in this gene result in autosomal recessive Charcot-Marie-Tooth disease type 4C, a childhood-onset neurodegenerative disease characterized by demyelination of motor and sensory neurons. This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 881 of the SH3TC2 protein (p.Asn881Ser). This variant is present in population databases (rs80338930, gnomAD 0.004%). This missense change has been observed in individual(s) with autosomal recessive Charcot Marie Tooth disease (PMID: 16924012, 21291453). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 377021). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on SH3TC2 protein function. Experimental studies have shown that this missense change affects SH3TC2 function (PMID: 20028792, 20826437, 23553667). For these reasons, this variant has been classified as Pathogenic. -
