Mutations in this gene result in autosomal recessive Charcot-Marie-Tooth disease type 4C, a childhood-onset neurodegenerative disease characterized by demyelination of motor and sensory neurons. Variant summary: SH3TC2 c.2528G>T (p.Gly843Val) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251128 control chromosomes. c.2528G>T has been reported in the literature in the presumed compound heterozygous and homozygous states in multiple individuals affected with clinical features of Charcot-Marie Disease Type 4C (example, Rehbein_2023, Volodarsky_2021). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 36947133, 32376792). ClinVar contains an entry for this variant (Variation ID: 476899).
