Same nucleotide change resulting in same amino acid change has been previously reported to be associated with SLC26A2 related disorder (PMID: 26077908). Therefore, this variant is classified as Likely pathogenic according to the recommendation of ACMG/AMP guideline. -

This sequence change replaces aspartic acid with valine at codon 673 of the SLC26A2 protein (p.Asp673Val). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and valine. This variant is present in population databases (rs774700553, ExAC 0.01%). This missense change has been observed in individual(s) with SLC26A2-related conditions (PMID: 26077908). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
