Loss-of-function variants in SQSTM1 are known to be pathogenic (PMID: 27545679, 29959261). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individuals with autosomal recessive neurodegeneration with ataxia, dystonia, and gaze palsy (NADGP) (PMID: 27545679).
