SDHA (HGNC:10680): (succinate dehydrogenase complex flavoprotein subunit A) This gene encodes a major catalytic subunit of succinate-ubiquinone oxidoreductase, a complex of the mitochondrial respiratory chain. The complex is composed of four nuclear-encoded subunits and is localized in the mitochondrial inner membrane. Mutations in this gene have been associated with a form of mitochondrial respiratory chain deficiency known as Leigh Syndrome. A pseudogene has been identified on chromosome 3q29. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Another alteration impacting the same donor site (c.1663+3G>C) has been detected in multiple individuals with a paraganglioma or gastrointestinal stromal tumor (Dwight T et al. Am J Surg Pathol, 2013 Feb;37:226-33; Ben Aim L et al. J Med Genet, 2019 08;56:513-520). c.1663+1G>A is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD).
