Variant summary: ARSB c.1442C>T (p.Pro481Leu) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251428 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1442C>T has been reported in the literature in a compound heterozygous individual affected with halitosis, slight photophobia, dyslexia, and mild hearing loss but not with typical presentations of Mucopolysaccharidosis Type VI (Maroteaux-Lamy Syndrome) (Brooks_2005). These report(s) do not provide unequivocal conclusions about association of the variant with Mucopolysaccharidosis Type VI (Maroteaux-Lamy Syndrome). At least one publication reports experimental evidence evaluating an impact on protein function. These results showed comparable specific activity of the mutant protein compared to that of wild type protein (Brooks_2005).
