This variant is present in population databases (rs781977997, gnomAD 0.007%). This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 455 of the LAMA4 protein (p.Arg455Gln). -

A variant of uncertain significance has been identified in the LAMA4 gene. The R455Q variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is observed in 1/10338 (0.010%) alleles from individuals of African ancestry and 4/66320 (0.006%) alleles from individuals of Non-Finnish European ancestry in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R455Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved in mammals. Nevertheless, in silico analysis predicts this variant likely does not alter the protein structure/function. -

The c.1364G>A (p.R455Q) alteration is located in exon 12 (coding exon 11) of the LAMA4 gene. This alteration results from a G to A substitution at nucleotide position 1364, causing the arginine (R) at amino acid position 455 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
