This alteration results from a A to G substitution at nucleotide position 3527, causing the histidine (H) at amino acid position 1176 to be replaced by an arginine (R). This sequence change replaces histidine, which is basic and polar, with arginine, which is basic and polar, at codon 1176 of the TBC1D32 protein (p.His1176Arg). This variant is present in population databases (rs757868336, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with TBC1D32-related conditions. ClinVar contains an entry for this variant (Variation ID: 2547813). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
