Variant summary: COL11A2 c.5071-7C>G alters a nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: One predict the variant abolishes a 3' acceptor site. Three predict the variant weakens a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 9e-05 in 244202 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in COL11A2 causing COL11A2-Related Disorders, allowing no conclusion about variant significance. To our knowledge, no occurrence of c.5071-7C>G in individuals affected with COL11A2-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported.
