Genes affected
LHFPL5 (HGNC:21253): (LHFPL tetraspan subfamily member 5) This gene is a member of the lipoma HMGIC fusion partner (LHFP) gene family, which is a subset of the superfamily of tetraspan transmembrane protein encoding genes. Mutations in this gene result in deafness in humans, and a mutation in a similar gene in mice results in deafness and vestibular dysfunction with severe degeneration of the organ of Corti. It is proposed to function in hair bundle morphogenesis. [provided by RefSeq, Jul 2008]

Uncertain significance, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineSep 25, 2018The p.Arg159Cys variant in LHFPL5 has been identified by our laboratory in 1 ind ividual with hearing loss. It has also been identified in 0.03% (10/30782) of So uth Asian chromosomes by gnomAD (http://gnomad.broadinstitute.org). Computationa l prediction tools and conservation analyses suggest that the p.Arg159Cys varian t may impact the protein, though this information is not predictive enough to de termine pathogenicity. In summary, the clinical significance of the p.Arg159Cys variant is uncertain. ACMG/AMP Criteria applied: PM2_Supporting, PP3. -
