POLH (HGNC:9181): (DNA polymerase eta) This gene encodes a member of the Y family of specialized DNA polymerases. It copies undamaged DNA with a lower fidelity than other DNA-directed polymerases. However, it accurately replicates UV-damaged DNA; when thymine dimers are present, this polymerase inserts the complementary nucleotides in the newly synthesized DNA, thereby bypassing the lesion and suppressing the mutagenic effect of UV-induced DNA damage. This polymerase is thought to be involved in hypermutation during immunoglobulin class switch recombination. Mutations in this gene result in XPV, a variant type of xeroderma pigmentosum. Studies have shown that disruption of this splice site is associated with altered splicing resulting in multiple RNA products (PMID: 27004399). Disruption of this splice site has been observed in individuals with xeroderma pigmentosum (PMID: 18368133, 27004399, 27664908). It has also been observed to segregate with disease in related individuals. This sequence change affects a donor splice site in intron 6 of the POLH gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in POLH are known to be pathogenic (PMID: 11773631, 24130121, 25256075). This variant is not present in population databases (gnomAD no frequency). - |
