The c.8420G>A (p.R2807H) alteration is located in exon 57 (coding exon 56) of the COL12A1 gene. This alteration results from a G to A substitution at nucleotide position 8420, causing the arginine (R) at amino acid position 2807 to be replaced by a histidine (H). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00011 in 249284 control chromosomes (gnomAD). The observed variant frequency is approximately 108 fold of the estimated maximal expected allele frequency for a pathogenic variant in COL12A1 causing Bethlem myopathy 2 phenotype (1e-06). To our knowledge, no occurrence of c.8420G>A in individuals affected with Bethlem myopathy 2 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 542511). Based on the evidence outlined above, the variant was classified as likely benign. -
