Arrhythmogenic cardiomyopathy with wooly hair and keratoderma Uncertain:1
This missense variant replaces threonine with methionine at codon 356 of the DSP protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two unrelated individuals affected with arrhythmogenic cardiomyopathy (PMID: 36431211, 37936624), in two homozygous siblings affected with dilated cardiomyopathy, and in their consanguineous parents who were unaffected heterozygotes (PMID: 36672924). This variant has been identified in 3/251198 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with arrhythmogenic left ventricular cardiomyopathy (PMID: 36431211), in two homozygous siblings affected with dilated cardiomyopathy, and in their consanguineous parents who were unaffected heterozygotes (PMID: 36672924). Cardiovascular phenotype Uncertain:1
The p.T356M variant (also known as c.1067C>T), located in coding exon 9 of the DSP gene, results from a C to T substitution at nucleotide position 1067. The threonine at codon 356 is replaced by methionine, an amino acid with similar properties. This alteration has been reported in arrhythmogenic right ventricular cardiomyopathy (ARVC) cohorts; however, clinical details were limited (Marschall C et al. Cardiovasc Diagn Ther, 2019 Oct;9:S292-S298; Bariani R et al. J Clin Med, 2022 Nov;11:[ePub ahead of print]; Maurer C et al. Genes (Basel), 2023 Jan;14:[ePub ahead of print]). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
