Variant summary: DSP c.4973C>T (p.Ser1658Phe) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 250690 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.4973C>T has been reported in the literature in at least one individual affected with arrhythmogenic right ventricular cardiomyopathy without evidence for causality (e.g. Rigato_2013). This report does not provide unequivocal conclusions about association of the variant with Arrhythmogenic Right Ventricular Dysplasia/Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 24070718). ClinVar contains an entry for this variant (Variation ID: 180332). The DSP c.4973C>T variant is predicted to result in the amino acid substitution p.Ser1658Phe. This variant was reported in the compound heterozygous state in two patients from an arrhythmogenic right ventricular cardiomyopathy (ARVC) cohort (Rigato et al. 2013. PubMed ID: 24070718). In a separate study assessing the pathogenicity and penetrance of various ARVC-related genes, the p.Ser1658Phe was listed as "questioned." (Ye et al. 2019. PubMed ID: 31402444). This variant is reported in 0.00088% of alleles in individuals of European (Non-Finnish) descent in gnomAD (http://gnomad.broadinstitute.org/variant/6-7581396-C-T). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
