Genes affected
MYO6 (HGNC:7605): (myosin VI) This gene encodes a reverse-direction motor protein that moves toward the minus end of actin filaments and plays a role in intracellular vesicle and organelle transport. The protein consists of a motor domain containing an ATP- and an actin-binding site and a globular tail which interacts with other proteins. This protein maintains the structural integrity of inner ear hair cells and mutations in this gene cause non-syndromic autosomal dominant and recessive hearing loss. Alternative splicing results in multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Jul 2014]

Uncertain significance, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Apr 19, 2017 | Variant classified as Uncertain Significance - Favor Benign. The p.Met18Val vari ant in MYO6 has not been previously reported in individuals with hearing loss, b ut has been identified in 0.14% (15/10404) of African chromosomes by the Exome A ggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs142516093). Although this variant has been seen in the general population, its frequency i s not high enough to rule out a pathogenic role. Computational prediction tools suggest that the p.Met18Val variant may not impact the protein, though this info rmation is not predictive enough to rule out pathogenicity. In summary, the clin ical significance of the p.Met18Val variant is uncertain. - |

Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jul 06, 2021 | In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C0"). ClinVar contains an entry for this variant (Variation ID: 504641). This variant has not been reported in the literature in individuals affected with MYO6-related conditions. This variant is present in population databases (rs142516093, ExAC 0.1%). This sequence change replaces methionine with valine at codon 18 of the MYO6 protein (p.Met18Val). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and valine. - |
