The p.I155M variant (also known as c.465C>G), located in coding exon 4 of the RINT1 gene, results from a C to G substitution at nucleotide position 465. The isoleucine at codon 155 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear. -

This sequence change replaces isoleucine with methionine at codon 155 of the RINT1 protein (p.Ile155Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. This variant is present in population databases (rs79893877, ExAC 0.001%). This variant has not been reported in the literature in individuals with RINT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0").
