Genes affected
RINT1 (HGNC:21876): (RAD50 interactor 1) This gene encodes a protein first identified for its ability to interact with the RAD50 double strand break repair protein, with the resulting interaction implicated in the regulation of cell cycle progression and telomere length. The encoded protein may also play a role in trafficking of cellular cargo from the endosome to the trans-Golgi network. Mutations in this gene may be associated with breast cancer in human patients. Uncertain significance, criteria provided, single submitterclinical testingInvitaeDec 07, 2019In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with RINT1-related disease. ClinVar contains an entry for this variant (Variation ID:Ā¬ā€ 410795). This variant is present in population databases (rs747465384, ExAC 0.001%). This sequence change replaces glutamine with arginine at codon 289 of the RINT1 protein (p.Gln289Arg). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and arginine. -
