The p.S647L variant (also known as c.1940C>T), located in coding exon 13 of the RINT1 gene, results from a C to T substitution at nucleotide position 1940. The serine at codon 647 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 647 of the RINT1 protein (p.Ser647Leu).
