This variant is present in population databases (rs540008835, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with SLC26A4-related conditions. Variant summary: SLC26A4 c.929C>T (p.Ala310Val) results in a non-conservative amino acid change located in the SLC26A/SulP transporter domain (IPR011547) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.6e-05 in 251038 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.929C>T has been reported in the literature in an individual affected with Congenital Hypothyroidism (de Filippis_2017). These report(s) do not provide unequivocal conclusions about association of the variant with Pendred Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 28444304). ClinVar contains an entry for this variant (Variation ID: 558589).
