The p.D44V variant (also known as c.131A>T), located in coding exon 2 of the CFTR gene, results from an A to T substitution at nucleotide position 131. The aspartic acid at codon 44 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
This sequence change replaces aspartic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 44 of the CFTR protein (p.Asp44Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of cystic fibrosis (PMID: 1379210). This variant is also known as 263A>T. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CFTR protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
