This nonsense variant (c.327T>G) on exon 4 of CFTR results in premature termination (p.Tyr109Ter) expected to result in loss of function (PVS1). This variant is not found in gnomAD databases (PM2) and several computational models predict pathogenicity (PP3). An alternate allele (codon TAA), c.327T>A (p.Tyr109Ter) is reported as pathogenic in Feuillet-Fieux et al., PMID 15025720. We interpret c.327T>G to be likely pathogenic. -
