This variant is present in population databases (rs397508359, gnomAD 0.1%). This variant has not been reported in the literature in individuals affected with CFTR-related conditions. ClinVar contains an entry for this variant (Variation ID: 53467). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The methionine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
The p.T760M variant (also known as c.2279C>T), located in coding exon 14 of the CFTR gene, results from a C to T substitution at nucleotide position 2279. The threonine at codon 760 is replaced by methionine, an amino acid with similar properties. This variant was detected in a cohort of individuals diagnosed with cystic fibrosis in Jordan; however, specific genotype and phenotype information was not provided (Rawashdeh M et al. Ann Trop Paediatr, 2000 Dec;20:283-6). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Variant summary: CFTR c.2279C>T (p.Thr760Met) results in a non-conservative amino acid change located in the CFTR regulator domain (IPR025837) of the encoded protein sequence. Three of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 9.7e-05 in 236864 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in CFTR causing Cystic Fibrosis (9.7e-05 vs 0.013), allowing no conclusion about variant significance. c.2279C>T has been reported in the literature in individuals affected with Cystic Fibrosis or undergoing screening for the condition without strong evidence of causality (Bozdogan_2021, Ni_2022, Rawashdeh_2000, Al-Sadeq_2019). These reports do not provide unequivocal conclusions about association of the variant with Cystic Fibrosis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 33572515, 35313924, 11219165, 33374015, 30419605).
