This sequence change replaces serine with alanine at codon 977 of the CFTR protein (p.Ser977Ala). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and alanine. This variant is present in population databases (rs137975784, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with CFTR-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant disrupts the p.Ser977 amino acid residue in CFTR. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 19318346, 23361109, 23891399). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
