Specifically, this protein functions as a member of a multi-protein complex that binds to the TTAGGG repeats of telomeres, regulating telomere length and protecting chromosome ends from illegitimate recombination, catastrophic chromosome instability, and abnormal chromosome segregation. Increased transcriptional expression of this gene is associated with stomach carcinogenesis and its progression. Alternatively spliced transcript variants have been described. The isoleucine at codon 455 is replaced by valine, an amino acid with highly similar properties. This alteration has been identified in multiple cases of familial melanoma (Shi J et al. Nat Genet, 2014 May;46:482-6; Goldstein AM et al. JAAD Int, 2023 Jun;11:43-51). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
