The p.I616M variant (also known as c.1848C>G), located in coding exon 12 of the FLNC gene, results from a C to G substitution at nucleotide position 1848. The isoleucine at codon 616 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces isoleucine with methionine at codon 616 of the FLNC protein (p.Ile616Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. This variant is not present in population databases (ExAC no frequency).
