These filamin proteins crosslink actin filaments into orthogonal networks in cortical cytoplasm and participate in the anchoring of membrane proteins for the actin cytoskeleton. Three functional domains exist in filamin: an N-terminal filamentous actin-binding domain, a C-terminal self-association domain, and a membrane glycoprotein-binding domain. Mutations in this gene are a cause of cardiopathy. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2022]

The glycine at codon 2011 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been reported in individuals with restrictive cardiomyopathy (RCM) (Kolokotronis K et al. J Clin Med, 2020 Jul;9:[ePub ahead of print]; Lopes LR et al. Eur Heart J, 2021 Aug;42:3063-3073). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
