PcG family members form multimeric protein complexes, which are involved in maintaining the transcriptional repressive state of genes over successive cell generations. This protein associates with the embryonic ectoderm development protein, the VAV1 oncoprotein, and the X-linked nuclear protein. This protein may play a role in the hematopoietic and central nervous systems. Multiple alternatively splcied transcript variants encoding distinct isoforms have been identified for this gene. The variant is absent in the gnomAD Exomes. This variant causes a frameshift starting with codon Glycine 743, changes this amino acid to Alanine residue, and creates a premature Stop codon at position 20 of the new reading frame. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing Tatton-Brown et al., 2011. However, since this variant is present in the last exon and there are no other downstream variants reported functional studies will be required to prove protein truncation. For these reasons, this variant has been classified as Uncertain Significance. -
