This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 985 of the KCNH2 protein (p.Asn985Ser). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This missense change has been observed in individual(s) with Brugada syndrome (PMID: 16043162). ClinVar contains an entry for this variant (Variation ID: 67456). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on KCNH2 function (PMID: 16043162).
