Genes affected
SNX10 (HGNC:14974): (sorting nexin 10) This gene encodes a member of the sorting nexin family. Members of this family contain a phox (PX) domain, which is a phosphoinositide binding domain, and are involved in intracellular trafficking. This protein does not contain a coiled coil region, like some family members. This gene may play a role in regulating endosome homeostasis. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2010]

Pathogenic, criteria provided, single submitterclinical testingInvitaeAug 09, 2023This sequence change creates a premature translational stop signal (p.Arg16*) in the SNX10 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SNX10 are known to be pathogenic (PMID: 23123320, 23280965, 25811986). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. ClinVar contains an entry for this variant (Variation ID: 139565). This premature translational stop signal has been observed in individuals with infantile osteopetrosis (PMID: 23123320, 23280965). This variant is present in population databases (rs587777490, gnomAD 0.005%). -
