The encoded protein was found in a complex with other proteins that have a role in HR-DSBR. Knockdown of this gene reduced homologous recombination, and mutations in this gene were found in patients with spastic paraplegia. This sequence change replaces serine, which is neutral and polar, with tryptophan, which is neutral and slightly polar, at codon 589 of the AP5Z1 protein (p.Ser589Trp). This variant is present in population databases (no rsID available, gnomAD no frequency). This variant has not been reported in the literature in individuals affected with AP5Z1-related conditions. ClinVar contains an entry for this variant (Variation ID: 2165442). This alteration results from a C to G substitution at nucleotide position 1766, causing the serine (S) at amino acid position 589 to be replaced by a tryptophan (W).
