This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 962 of the EGFR protein (p.Arg962Gly). This variant is present in population databases (rs17337451, gnomAD 0.04%). This variant has not been reported in the literature in individuals affected with EGFR-related conditions. ClinVar contains an entry for this variant (Variation ID: 956413). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on EGFR protein function. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.

The p.R962G variant (also known as c.2884C>G), located in coding exon 24 of the EGFR gene, results from a C to G substitution at nucleotide position 2884. The arginine at codon 962 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on data from gnomAD, the frequency for this variant is above the maximum credible frequency for a disease-causing variant in this gene based on internally established thresholds (Karczewski et al. Nature. 2020 May;581(7809):434-443; Whiffin et al. Genet Med. 2017 10;19:1151-1158). Based on the supporting evidence, the association of this alteration with an increased risk of EGFR-related neonatal inflammatory skin and bowel disease is unknown; however, the association of this alteration with an increased risk of EGFR-related lung cancer is unlikely.
