The glycine at codon 218 is replaced by arginine, an amino acid with dissimilar properties. This variant has been reported in an individual with ovarian cancer diagnosed at age 41 and who had a family history of kidney and ovarian cancer (Kraus C et al. International Journal of Cancer, 2017 Jan;140:95-102). This amino acid position is poorly conserved in available vertebrate species.
