Genes affected
HSPB1 (HGNC:5246): (heat shock protein family B (small) member 1) This gene encodes a member of the small heat shock protein (HSP20) family of proteins. In response to environmental stress, the encoded protein translocates from the cytoplasm to the nucleus and functions as a molecular chaperone that promotes the correct folding of other proteins. This protein plays an important role in the differentiation of a wide variety of cell types. Expression of this gene is correlated with poor clinical outcome in multiple human cancers, and the encoded protein may promote cancer cell proliferation and metastasis, while protecting cancer cells from apoptosis. Mutations in this gene have been identified in human patients with Charcot-Marie-Tooth disease and distal hereditary motor neuropathy. [provided by RefSeq, Aug 2017]

ACMG classification
Classification made for transcript
Verdict is Likely_pathogenic. Variant got 8 ACMG points.
PVS1
Loss of function variant, product does not undergo nonsense mediated mRNA decay. Variant is located in the 3'-most exon, not predicted to undergo nonsense mediated mRNA decay. There are 8 pathogenic variants in the truncated region.
PM2
Very rare variant in population databases, with high coverage;
PP5
Variant 7-76304087-G-T is Pathogenic according to our data. Variant chr7-76304087-G-T is described in ClinVar as [Likely_pathogenic]. Clinvar id is 465276.Status of the report is criteria_provided_single_submitter, 1 stars.

Charcot-Marie-Tooth disease axonal type 2F Pathogenic:1
Likely pathogenic, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Apr 17, 2017 | A different missense variant downstream of this variant (p.P182L) has been determined to be pathogenic (PMID: 15122254, 23728742, 28077174). This suggests that deletion of this region of the HSPB1 protein is causative of disease. In summary, this variant is a novel nonsense change that is expected to delete 28 amino acids of HSPB1 important for the protein function. This evidence indicates that the variant is pathogenic, but additional data is needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a HSPB1-related disease. This sequence change results in a premature translational stop signal in the last exon of the HSPB1 mRNA at codon 178 (p.Glu178*). While this is not anticipated to result in nonsense mediated decay, it is expected to delete the last 28 amino acids of the HSPB1 protein. - |
