The p.P619L variant (also known as c.1856C>T), located in coding exon 9 of the DNAAF5 gene, results from a C to T substitution at nucleotide position 1856. The proline at codon 619 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 619 of the DNAAF5 protein (p.Pro619Leu). This variant is present in population databases (rs755871359, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with DNAAF5-related conditions. ClinVar contains an entry for this variant (Variation ID: 525305).
