Variant summary: COL1A2 c.2968G>T (p.Ala990Ser) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-05 in 251398 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in COL1A2 causing Ehlers-Danlos syndrome, cardiac valvular type (4e-05 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.2968G>T in individuals affected with Ehlers-Danlos syndrome, cardiac valvular type or other COL1A2-related disorders and no experimental evidence demonstrating its impact on protein function have been reported.
