Ritscher-Schinzel syndrome;C1863704:Hereditary spastic paraplegia 8 Uncertain:1
In summary, this is a rare missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "possibly damaging"; Align-GVGD: "Class C0"). This variant has not been published in the literature and is present in population databases (rs199936194, 0.05%). This sequence change replaces isoleucine with threonine at codon 54 of the KIAA0196 protein (p.Ile54Thr). The isoleucine residue is moderately conserved and there is a moderate physicochemical difference between isoleucine and threonine.
