The PLEC p.Arg2143His variant was not identified in the literature but was identified in dbSNP (ID: rs372652727), ClinVar (classified as a VUS by EGL Genetics and Invitae) and LOVD 3.0 (classified as a VUS). The variant was also identified in control databases in 33 of 256488 chromosomes at a frequency of 0.000129 (Genome Aggregation Database Feb 27, 2017). This alteration results from a G to A substitution at nucleotide position 6497, causing the arginine (R) at amino acid position 2166 to be replaced by a histidine (H). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
