The p.E661K variant (also known as c.1981G>A), located in coding exon 12 of the RECQL4 gene, results from a G to A substitution at nucleotide position 1981. The glutamic acid at codon 661 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

This variant is present in population databases (rs775332504, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with RECQL4-related conditions. ClinVar contains an entry for this variant (Variation ID: 459363).
