The c.701C>G (p.S234C) alteration is located in exon 5 (coding exon 5) of the RECQL4 gene. This alteration results from a C to G substitution at nucleotide position 701, causing the serine (S) at amino acid position 234 to be replaced by a cysteine (C). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

This sequence change replaces serine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 234 of the RECQL4 protein (p.Ser234Cys). This variant is present in population databases (rs746487281, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with RECQL4-related conditions. ClinVar contains an entry for this variant (Variation ID: 459512). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
