The protein localizes to the endoplasmic reticulum (ER), and may recycle between the ER and ER-Golgi intermediate compartment. Mutations in this gene are associated with a disorder characterized by progressive epilepsy with cognitive disabilities (EPMR), which is a subtype of neuronal ceroid lipofuscinoses (NCL). This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 70 of the CLN8 protein (p.Arg70Cys). This variant is present in population databases (rs765097897, gnomAD 0.007%). This missense change has been observed in individual(s) with clinical features of neuronal ceroid lipofuscinosis (PMID: 31069529, 37074398). ClinVar contains an entry for this variant (Variation ID: 242453). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt CLN8 protein function with a positive predictive value of 95%. This variant disrupts the p.Arg70 amino acid residue in CLN8. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 21990111, 31130284; internal data).
